Product Summary

The Quantikine Human Caspase-1 Immunoassay is a 3 hour solid phase ELISA designed to measure Caspase-1 in cell culture supernates. It contains recombinant human Caspase-1 and antibodies raised against the recombinant factor. This immunoassay has been shown to accurately quantitate the recombinant factor. Results obtained measuring natural human Caspase-1 showed dose-response curves that were parallel to the standard curves obtained using the recombinant Caspase-1. These results indicate that this kit can be used to determine relative mass values for natural human Caspase-1.

Background: Caspase-1

Caspases are a family of cytosolic aspartate-specific cysteine proteases involved in the initiation and execution of apoptosis. They are expressed as latent zymogens and are activated by an autoproteolytic mechanism or by processing by other proteases (frequently other caspases). Human caspases can be subdivided into three functional groups: cytokine activation (caspase-1, -4, -5, and -13), apoptosis initiation (caspase-2, -8, -9, -and -10), and apoptosis execution (caspase-3, -6, and -7).

Caspases are regulated by a variety of stimili, including APAF1, CFLAR/FLIP, NOL3/ARC, and members of the inhibitor of apoptosis (IAP) family such as BIRC1/NAIP, BIRC2/cIAP-1, BIRC3/cIAP-2, BIRC4/XIAP, BIRC5/Survivin, and BIRC7/Livin. IAP activity is modulated by DIABLO/SMAC or PRSS25/HTRA2/Omi. Cell-permeable and irreversible peptide inhibitors are also available for different caspases.

Assay Procedure

Refer to the product for complete assay procedure.

Bring all reagents and samples to room temperature before use. It is recommended that all samples, standards, and controls be assayed in duplicate.
  1.   Prepare all reagents, standard dilutions, and samples as directed in the product insert.
  2.   Remove excess microplate strips from the plate frame, return them to the foil pouch containing the desiccant pack, and reseal.

  3. 50 µL Assay Diluent
  4.   Add 50 µL of Assay Diluent to each well.

  5. 100 µL Standard, Control, or Sample
  6.   Add 100 µL of Standard, control, or sample to each well. Cover with a plate sealer, and incubate at room temperature for 1.5 hours.
  7.   Aspirate each well and wash, repeating the process twice for a total of 3 washes.

  8. 100 µL Antiserum
  9.   Add 100 µL of Antiserum to each well. Cover with a new plate sealer, and incubate at room temperature for 30 minutes.
  10.   Aspirate and wash 3 times.

  11. 100 µL Conjugate
  12.   Add 100 µL of Conjugate to each well. Cover with a new plate sealer, and incubate at room temperature for 30 minutes.
  13.   Aspirate and wash 3 times.

  14. 200 µL Substrate Solution
  15. Add 200 µL Substrate Solution to each well. Incubate at room temperature for 20 minutes. PROTECT FROM LIGHT.

  16. 50 µL Stop Solution
  17. Add 50 µL of Stop Solution to each well. Read at 450 nm within 30 minutes. Set wavelength correction to 540 nm or 570 nm.
