    Interleukin-18 Binding Proteins (IL-18 BP) are secreted immunoglobulin superfamily proteins that bind IL-18 and prevent the activation of IL-18 R alpha. IL-18 BPa is the predominantly expressed isoform of four alternatively spliced IL-18 BP isoforms and has high affinity for IL-18. Neutralization of IL-18 by IL-18 BP blocks T cell infiltration into sites of inflammation, the production of pro-inflammatory cytokines, and Th17 responses. IL-18 BP expression increases upon inflammatory stimulation, and circulating levels are elevated in systemic lupus erythematosus (SLE), active Crohn’s disease, ulcerative colitis, alcoholic hepatitis, and chronic renal insufficiency.

    Assessment of IL-18 BPa in Cell Culture Supernates. Human umbilical vein endothelial cells (HUVECs) were unstimulated or stimulated with TNF-alpha (Catalog # 210-TA) and IFN-gamma (Catalog # 285-IF). Following treatment, IL-18 BPa secretion was assessed in conditioned media using (A) the Human IL-18 BPa Quantikine ELISA Kit (Catalog # DBP180) or (B) by Western blot using a Goat Anti-human IL-18 BPa Antigen Affinity-Purified Polyclonal Antibody (Catalog # AF119).

    DNA Methyltransferase 3B (DNMT3B) is an approximately 95-110 kDa enzyme that transfers a methyl group from S-Adenosyl-L-methionine to the 5 carbon position of cytosine within DNA, thereby producing 5-methylcytosine. Along with DNMT3A, DNMT3B establishes initial DNA methylation patterns during development while DNMT1 maintains DNA methylation during replication. DNMT3B is highly expressed in undifferentiated embryonic stem cells. Expression of DNMT3B decreases during differentiation and is detected at low levels in adult somatic cells.

    DNMT3B in the iPS2 Human Stem Cell Line

    DNMT3B in the iPS2 Human Stem Cell Line. iPS2 human induced pluripotent stem cells were cultured in the absence of growth factors for three days to stimulate random differentiation. DNMT3B was detected in immersion-fixed iPS2 human induced pluripotent stem cells using a Mouse Anti-Human DNMT3B Monoclonal Antibody (Catalog # MAB7646). The cells were stained with the NorthernLights™ (NL)557-conjugated Donkey Anti-Mouse IgG Secondary Antibody (Catalog # NL007; red) (A). Oct-3/4 was detected using a Goat Anti-Human Oct-3/4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1759) followed by the NL493-conjugated Donkey Anti-Goat IgG Secondary Antibody (Catalog # NL003; green) (B). The nuclei were counterstained with DAPI (blue). Immunoreactivity of DNMT3B was localized to the nuclei of undifferentiated cells and co-localized with the pluripotent marker Oct-3/4 (C).

    Casein Kinase 2 (CK2) is serine/threonine kinase that includes two catalytic subunits (CK2 alpha and/or CK2 alpha’) and 2 identical regulatory subunits (CK2 beta). It is known to phosphorylate more than 300 different substrates and has diverse roles in apoptosis, cell cycle regulation, and gene transcription. The new antibody was developed using recombinant human CK2 alpha Asp253-Gln391 as the immunogen. This protein, including the region of divergence between CK2 alpha and CK2 alpha’, shares 96% amino acid sequence identity with the mouse and rat CK2 orthologs.

    Detection of Casein Kinase 2 alpha by Western Blot and ICC. (A) Casein Kinase 2 alpha was detected by the Mouse Anti-Human Casein Kinase 2 alpha Monoclonal Antibody (Catalog # MAB7957) using Western blot in lysates from human, mouse, and rat cell lines: HeLa human cervical epithelial carcinoma, MOLT-4 human acute lymphoblastic leukemia, NIH 3T3 mouse embryonic fibroblast, C6 rat glioma, and Rat 2 rat embryonic fibroblast. (B) The antibody detects Casein Kinase 2 alpha by ICC in the nuclei of the HEK293 human embryonic kidney cell line. The cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (upper panel; Catalog # NL007) and counterstained with DAPI (lower panel).
