Anti-idiotype antibodies are antibodies that bind to the variable region of another antibody. Since anti-idiotype antibodies are specific for the variable region, they have become a useful tool in pharmacokinetic and immunogenicity studies.
R&D Systems offers multiple types of anti-idiotype antibodies. Anti-idiotype antibodies can be blocking or non-blocking. Using different types of anti-idiotypes allows more flexibility in assay development.
Our anti-idiotype antibodies are specifically designed to detect the biosimilar antibody it is targeting. We perform extensive in-house testing to confirm the specificity and functionality of our anti-idiotype antibodies. We perform bridging assays, flow cytometry, binding inhibition, and direct ELISA testing.
Bridging Assay
The anti-idiotype antibody is tested in a bridging assay. The anti-idiotype antibody is used to capture and detect the biosimilar.
Flow Cytometry
The anti-idiotype antibody binds to the biosimilar antibody bound to its ligand on the cell (filled histogram). In the absence of the biosimilar antibody, there is no binding of the anti-idiotype antibody to the cell (open histogram).
Binding Inhibition
The blocking anti-idiotype antibody binds to the biosimilar antibody so it cannot bind to its ligand on the cell (orange histogram). In the absence of the blocking anti-idiotype antibody, the biosimilar antibody can bind to the cell (black dotted line). An isotype control to the anti-idiotype antibody is used to show that binding is only blocked by the anti-idiotype antibody (blue line).
Direct ELISA
Direct ELISA testing is used to determine specificity. Biosimilars are coated on a plate. The anti-idiotype antibody is used to detect the biosimilar antibody but does not detect other irrelevant biosimilar antibodies.
