Scientific Data
Detection of Human AIM2 by Western Blot. Western blot shows lysates of Daudi human Burkitt's lymphoma cell line. PVDF membrane was probed with 2 µg/mL of Rabbit Anti-Human AIM2 Monoclonal Antibody (Catalog # MAB9965) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for AIM2 at approximately 40 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: AIM2
Human Interferon-inducible protein AIM2 (also known as absent in melanoma 2 or simply AIM2), encoded by the AIM2 gene, is a 343 amino acids (aa), 39 KDa protein with a N-terminal DAPIN (or pyrin) domain (amino acids 1-87) and a C-terminal HIN-200 domain (amino acids 138-337), which is known to have two oligonucleotide-binding folds. Recent research has shown that AIM2 is part of the inflammasome and is involved in innate immune response by recognizing cytosolic double-stranded DNA and inducing caspase-1-activating the inflammasome in macrophages. Upon binding to DNA, Aim-2 undergoes oligomerization and associates with PYCARD initiating the recruitment of caspase-1 precursors and processing of IL-1b and IL-18. Over aa 71-180, human Aim2 shares 90% aa sequence identity with macaque Aim2.
