Detection of Human alpha 1‑Acid Glycoprotein by Western Blot. Western blot shows human plasma. PVDF membrane was probed with 0.1 µg/mL of Goat Anti-Human a1-Acid Glycoprotein Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3694) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for a1-Acid Glycoprotein at approximately 45-50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Detection of Human alpha 1‑Acid Glycoprotein by Simple WesternTM. Simple Western lane view shows lysates of human serum, loaded at 1:5000. A specific band was detected for a1-Acid Glycoprotein at approximately 79 kDa (as indicated) using 5 µg/mL of Goat Anti-Human a1-Acid Glycoprotein Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3694) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: alpha 1-Acid Glycoprotein
alpha 1-acid Glycoprotein, also known as orosomucoid-1, is a positive acute-phase reactant that is produced by the liver and secreted in plasma. Plasma alpha 1-acid concentration increases in response to inflammation and infection. alpha 1-acid glycoprotein belongs to the lipocalin family and binds numerous basic and neutral lipophilic drugs and steroid hormones. It is a 183 amino acid protein with 5 N-linked glycans that comprises 45% of the 43 kDa mass of the protein. The human and mouse protein share 43% amino acid sequence identity.
