Sample Values

Serum/Plasma - Samples from apparently healthy volunteers were evaluated for the presence of human Angiopoietin-2 in this assay. No medical histories were available for the donors used in this study.

Sample TypeMean (pg/mL)Range (pg/mL)Standard Deviation (pg/mL)
Serum (n=60)24941065-89071341
EDTA plasma (n=35)19641071-4389808
Heparin plasma (n=35)20491009-4973913

Saliva - Nine samples were evaluated for the presence of human Angiopoietin-2 in this assay and ranged from 247-822 pg/mL.

Cell Culture Supernates - Two cultures of HUVEC human umbilical vein endothelial cells were grown to confluency in EGM supplemented with 2% fetal bovine serum, 2 mM L-glutamine, 100 U/mL penicillin, 100 μg/mL streptomycin sulfate and bovine brain extract. Aliquots of the cell culture supernates were removed and assayed for levels of human Angiopoietin-2.

SampleValues (pg/mL)
HUVEC Culture 12326
HUVEC Culture 27611

Note: Significant levels of Angiopoietin-2 are found in fetal bovine, bovine, porcine, equine, and rabbit serum. The background level of Angiopoietin-2 in control medium should be determined and subtracted from samples of conditioned medium.

Product Summary

The Quantikine Human Angiopoietin-2 Immunoassay is a 4.5 hour solid-phase ELISA designed to measure human Angiopoietin-2 in cell culture supernates, serum, plasma, and saliva. It contains NS0-expressed recombinant human Angiopoietin-2 and has been shown to accurately quantitate the recombinant factor. Results obtained using natural human Angiopoietin-2 showed linear curves that were parallel to the standard curves obtained using the Quantikine kit standards. These results indicate that this kit can be used to determine relative mass values for naturally occurring human Angiopoietin-2.

Recovery

The recovery of Angiopoietin-2 spiked to levels throughout the range of the assay in various matrices was evaluated.

Sample Type Average % Recovery Range %
Cell Culture Media (n=4) 103 98-113
EDTA Plasma (n=4) 95 85-104
Heparin Plasma (n=4) 94 87-104
Serum (n=4) 100 90-107

Linearity

To assess the linearity of the assay, samples containing and/or spiked with high concentrations of Angiopoietin-2 were serially diluted with Calibrator Diluent to produce samples with values within the dynamic range of the assay.

Background: Angiopoietin-2

Angiopoietins, Ang-1, Ang-2, and Ang-3 (mouse)/Ang-4 (human), are natural agonists or antagonists of the Tie-2 receptor tyrosine kinase and are important modulators of angiogenesis. Two domains characterize the angiopoietin family of proteins: an N-terminal coiled-coil domain that mediates homo-oligomerization, and a C-terminal fibrinogen-like domain that binds Tie-2.

Assay Procedure

Refer to the product for complete assay procedure.

Bring all reagents and samples to room temperature before use. It is recommended that all samples, standards, and controls be assayed in duplicate.
  1.   Prepare all reagents, standard dilutions, and samples as directed in the product insert.
  2.   Remove excess microplate strips from the plate frame, return them to the foil pouch containing the desiccant pack, and reseal.

  3. 100 µL Assay Diluent
  4.   Add 100 µL of Assay Diluent to each well.

  5. 50 µL Standard, Control, or Sample
  6.   Add 50 µL of Standard, control, or sample to each well. Cover with a plate sealer, and incubate at room temperature for 2 hours on a horizontal orbital microplate shaker.
  7.   Aspirate each well and wash, repeating the process 3 times for a total of 4 washes.

  8. 200 µL Conjugate
  9.   Add 200 µL of Conjugate to each well. Cover with a new plate sealer, and incubate at room temperature for 2 hours on the shaker.
  10.   Aspirate and wash 4 times.

  11. 200 µL Substrate Solution
  12.   Add 200 µL Substrate Solution to each well. Incubate at room temperature for 30 minutes on the benchtop. PROTECT FROM LIGHT.

  13. 50 µL Stop Solution
  14.   Add 50 µL of Stop Solution to each well. Read at 450 nm within 30 minutes. Set wavelength correction to 540 nm or 570 nm.

Human Angiopoietin-2 Quantikine ELISA Kit
By Anonymous on 03/24/2022
Sample Tested: Plasma,Serum,Urine

Urine was very low concentration & mostly undetectable at 2-fold. Plasma & Serum showed pretty good linearity up to 20-fold dilution. Almost no matrix effect in plasma at 20-fold dilution.
