Detection of Human Apolipoprotein B100/ApoB100 by Western Blot. Western blot shows human plasma. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human Apolipoprotein B100/ApoB100 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3260) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for Apolipoprotein B100/ApoB100 at approximately 500 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Detection of Human Apolipoprotein B100/ApoB100 by Simple WesternTM. Simple Western shows lysates of human plasma, loaded at 1:50. A specific band was detected for Apolipoprotein B100/ApoB100 at approximately 307 kDa (as indicated) using 5 µg/mL of Goat Anti-Human Apolipoprotein B100/ApoB100 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3260). This experiment was conducted under reducing conditions and using the 66-440kDa separation system.

Background: Apolipoprotein B100/ApoB100

ApoB100 is a major serum Apolipoprotein synthesized by the liver. It is an important constituent of LDL, VLDL and IDL particles. It interacts with the LDL receptor and is involved in the delivery of cholesterol to tissues. Within the peptide sequence of the immunogen, human and mouse ApoB100 share 74% amino acid sequence identity. The sequence within the immunogen is absent in ApoB48, an alternative splice isoform that is exclusively synthesized by the gut. ApoB48 contains the amino terminal 47% of ApoB100 and lacks the LDL receptor region.
