Scientific Data
Detection of Human Desmoplakin by Western Blot. Western blot shows lysate of COLO 205 human colorectal adenocarcinoma cell line. PVDF membrane was probed with 0.5 µg/mL of Mouse Anti-Human Desmoplakin Monoclonal Antibody (Catalog # MAB9080) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). Specific bands were detected for Desmoplakin at approximately 260 and 330 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Desmoplakin
DSP
(desmoplakin) is the major high molecular weight protein of desmosomes, the
most common type of intercellular junction in vertebrate epithelial cells. DSP is involved in the organization of the
desmosomal cadherin-plakoglobin complex, as well as in the anchoring of
intermediate filaments to desmosomes.
These functions are thought to impart tensile strength and integrity to
epithelial and vascular tissues.
Mutations in DSP result in several clinical phenotypes, ranging from
mild skin conditions, to severe skin blistering with subsequent dehydration, to
lethal heart defects. Two major
desmoplakin isoforms, DSPI and DSPII, are encoded by alternative mRNA
transcripts differentially spliced from the same gene. Human DSPI is 2,871 amino acids (aa) in length,
while DSPII lacks part of a
central alpha-helical rod domain, and is 599 aa
shorter.
