Background: ICAT
Human ICAT is a physiological inhibitor of the beta -Catenin/Wnt signaling pathway. It is 9 kDa in size and found in both cytosol and nucleus. The molecule contains an alpha ‑helical N-terminal region (aa 9‑54) and an acidic, extended C-terminal region (aa 55‑78). The alpha -helical region binds to beta -Catenin, displacing CBP/p300, while the acidic C-terminus interacts with the beta -Catenin N-terminus, blocking Tcf interaction. Human ICAT shares 97% aa sequence identity with mouse and bovine ICAT, respectively.
