Detection of Mouse IkB-beta by Western Blot. Western blot shows lysates of NIH-3T3 mouse embryonic fibroblast cell line, BaF3 mouse pro-B cell line, M1 mouse myeloid leukemia cell line. PVDF membrane was probed with 1 µg/mL of Human/Mouse IkB-beta Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5225) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for IkB-beta at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: IkB-beta
I kappa B-beta (I kappa B-beta; also NF-kappa-BIB) is a 46 kDa member of the I kappa B-beta inhibitor family of proteins. It is highly expressed in testicular spermatids, and serves to block NF-kappa B activity. This is accomplished by I kappa B-beta binding to either an NF-kappa B dimer, or a kappa B-Ras:NF-kappa B dimer complex. Mouse I kappa B-beta is 359 amino acids (aa) in length it contains a signal response region (aa 1‑50), six ankyrin repeats (aa 54‑308) and a PEST domain whose phosphorylation at Ser312 and Ser314 blocks NF‑ kappa B activation. Phosphorylation at Ser19 and Ser23 promotes I kappa B‑ beta ubiquitination and degradation. Full‑length mouse I kappa B‑ beta shares 94% and 84% aa identity with rat and human I kappa B-beta, respectively.
