Detection of Rat Neuroligin 3/NLGN3 by Western Blot. Western blot shows lysates of rat embryonic cortical neurons. PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human/Mouse/Rat Neuroligin 3/NLGN3 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6088) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Neuroligin 3/NLGN3 at approximately 117 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.

Neuroligin 3/NLGN3 in Human Brainstem. Neuroligin 3/NLGN3 was detected in immersion fixed paraffin-embedded sections of human brainstem using Sheep Anti-Human/Mouse/Rat Neuroligin 3/NLGN3 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6088) at 10 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). Background: Neuroligin 3/NLGN3

Neuroligin 3 (NLGN3 or NL3) is an 100-120 kDa type I transmembrane glycoprotein that is a non-enzymatic member of the type-B carboxylesterase/lipase family. It is expressed on postsynaptic neurons and initiates excitatory presynapse maturation through calcium-dependent binding to a select isoform of beta -neurexin. The 848 amino acid (aa) human NLGN3 contains a 38 aa signal sequence and a 670 aa extracellular domain that shares 99.7% aa identity with mouse and rat NLGN3. Alternate splice forms of human NLGN3 encode 828, 808 and 711 aa lacking aa 153-172, 153-192, and 1-118 (plus 153-172), respectively. Certain NLGN3 mutations are associated with rare cases of autism.
