Detection of Human, Mouse, and Rat PP2C epsilon /PPM1L by Western Blot. Western blot shows lysates of BJAB human Burkitt's lymphoma cell line, Nb2-11 rat lymphoma cell line, Neuro-2A mouse neuroblastoma cell line, and L1.2 mouse pro-B cell line. PVDF membrane was probed with 0.5 µg/mL of Mouse Anti-Human PP2C e/PPM1L Monoclonal Antibody (Catalog # MAB6914) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for PP2C e/PPM1L at approximately 35 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: PP2C epsilon/PPM1L
PP2C epsilon, also known as PPM1L, is a 34 kDa (predicted) widely expressed Ser/Thr phosphatase. It consists of a 25 amino acid (aa) extracellular domain, a 17 aa transmembrane segment, and a 318 aa cytoplasmic region (aa 43-360) which contains the phosphatase domain (aa 91-344). Alternate splicing generates additional isoforms of human PP2C epsilon with 179 aa or 127 aa N-terminal truncations or a 166 aa C-terminal truncation. The PP2C epsilon -mediated dephosphorylation of TAK1 and ASK1 is regulated by oxidative stress and inflammatory cytokines. Loss of PP2C epsilon function is associated with the development of metabolic syndrome. Within aa 43-178, human PP2C epsilon shares 99% aa sequence identity with mouse and rat PP2C epsilon.
