Human p27/Kip1 IHC Detection Kit Summary

p27/Kip 1 IHC Detection Kit 

The cyclin-dependent kinase (cdk) inhibitor p27, also known as Kip1, is a tumor suppressor protein. Loss of p27 function has been linked to genetic instability and the progression of many types of cancer. R&D Systems’ Human p27 Immunohistochemistry Detection Kit (Catalog # CTS2256) includes all reagents required for excellent immunoreactivity and localization of the human p27 antigen in formalin-fixed tissues.

The antibody included detects p27 in human and mouse tissues. Specificity was confirmed by comparing staining on tissue sections of normal versus p27 knockout mice and by Western blot on human prostate tissue lysates. Heat-Induced Epitope Retrieval (HIER) pretreatment is required for the detection of this antigen in formalin-fixed, paraffin-embedded (FFPE) sections of human and mouse normal and cancer tissues.

Background: p27/Kip1

The cyclin-dependent kinase (cdk) inhibitor p27/Kip1 is a tumor suppressor protein that acts in the nucleus to enforce cell cycle checkpoints. p27/Kip1 binds to and inhibits many cyclin/cdk complexes, including cyclin D/cdk4, cyclin E/cdk2, and cyclin A/cdk2. This inhibition can subsequently block progression through different phases of the cell cycle. p27/Kip1 is phosphorylated at T198 in cells treated with the AMP-dependent protein kinase (AMPK) activator AICAR (5-amino-4-imidazolecarboxamide riboside) or glucose withdrawal.
