Detection of Human and Primate Ig Kappa Light Chain by Western Blot. Western blot shows human kappa light chain protein, human lambda light chain protein (negative control), human serum, primate serum, mouse serum (negative control), and rat serum (negative control). PVDF membrane was probed with 2 µg/mL of Rabbit Anti-Human/Primate Ig Kappa Light Chain Monoclonal Antibody (Catalog # MAB100502) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Ig Kappa Light Chain at approximately 25 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Ig Kappa Light Chain

The immunoglobulin light chain is the smaller subunit of an antibody and in humans can be expressed in two types: as a kappa ( kappa ) or a lambda ( lambda ) chain.  Antibodies are produced by B-Cells that are clonal and each expresses only one type of light chain. The light chain class remains fixed for the life of the B lymphocyte. The ratio of kappa and lambda light chains can be used to determine disease status by immunohistochemistry or ELISA based assays.
