Background: ATF2

ATF2 (activating transcription factor 2; also CRE-BP1) is a 60 kDa member of the ATF subfamily, basic-region leucine zipper family of DNA binding proteins. It has at least two activities. First, it binds to an octameric palindromic CRE (cAMP response element) 5’-TGACGTCA-3’, and secondary, it shows histone acetyltransferase (HAT) activity towards histones H2B and H4. Its palindromic binding activity depends upon homodimerization, or heterodimerization with either c-Jun or CRE-Bpa. Human ATF2 is 505 amino acids (aa) in length, and contains a C2H2-type zinc finger segment (aa 25-49), a HAT region (aa 112-350), and a basic leucine zipper domain (aa 350-414). There are over a dozen alternate splice forms that, in their entirety, impact all segments of the molecule. ATF2 is phosphorylated at multiple sites, including S62/S90/S112/S340/S368/S490/S498 and T69/T71. The Thr phosphorylations increase transcriptional and HAT activity. Serine phosphorylations at S490/S498 by ATM contribute to DNA damage repair after ionizing radiation. Human ATF2 is greater that 95% aa identical to mouse ATF2 and 100% identical to rat ATF2.
