Detection of Human SKAP55 by Western Blot. Western blot shows lysates of Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 1 µg/mL Goat Anti-Human SKAP55 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4264) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). For additional reference, recombinant human SKAP55 (1 ng) was included. A specific band for SKAP55 was detected at approximately 55 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: SKAP55
Src kinase-associated phosphoprotein of 55 kDa (SKAP55) is an adaptor protein expressed exclusively in mononuclear cells of hematopoietic lineage, with preferential expression in T lymphocytes and monocytes. Also known as SCAP1, SKAP55 regulates integrin-mediated adhesion and promotes synapse formation between T cells and antigen-presenting cells. SKAP55 is expressed as a constitutively tyrosine-phosphorylated protein that interacts with the SH2 domain of Fyn in resting cells.
