Detection of Human STAT2 by Western Blot. Western blot shows lysates of K562 human chronic myelogenous leukemia cell line and Daudi human Burkitt's lymphoma cell line. PVDF membrane was probed with 1 µg/mL of Rat Anti-Human STAT2 Monoclonal Antibody (Catalog # MAB16662) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for STAT2 at approximately 110 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Detection of Human STAT2 by Simple WesternTM. Simple Western lane view shows lysates of K562 human chronic myelogenous leukemia cell line and Daudi human Burkitt's lymphoma cell line, loaded at 0.2 mg/mL. A specific band was detected for STAT2 at approximately 110 kDa (as indicated) using 10 µg/mL of Rat Anti-Human STAT2 Monoclonal Antibody (Catalog # MAB16662) followed by 1:50 dilution of HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.
Western Blot Show Human STAT2 Specificity Using Knockout Cell Line. Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and STAT2 knockout HeLa cell line (KO). PVDF membrane was probed with 1 µg/mL of Rat Anti-Human STAT2 Monoclonal Antibody (Catalog # MAB16662) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for STAT2 at approximately 110 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: STAT2
Signal transducer and activator of transcription 2 (STAT2) is a member of the transcription factor STAT family with an apparent M.W. of 113 kDa in SDS-PAGE gels. Human STAT2 is 851 amino acids (aa) in length and contains one SH2 domain (aa 572-667). Splicing variants produce two isoforms for human STAT2. The short isoform has a 32 aa substitution and a 199 aa deletion corresponding to aa 621‑652 and aa 653‑851 in the long isoform, respectively. Human STAT2 shares 73% and 65% aa sequence identity with rat and mouse STAT2, in that order. STAT2 functions as a signal transducer and activator of transcription that mediates signaling by type I IFNs (IFN-alpha and IFN-beta ), and it is also required for myogenic differentiation.
