Build NowPorcine Luminex® Discovery Assay Summary
Assay Type | Magnetic bead-based multiplex assay for the Luminex® platform |
Format | 1 x 96-well microplate and Magnetic antibody-coated beads |
Species | Porcine |
Analytes Detected | Please see analyte list in assay customization tool below. |
Performance Validation | Luminex Assays are validated for use with cell culture supernatants, plasma and serum. All Luminex assays are tested for sensitivity, intra-assay precision, inter-assay precision and to ensure assay linearity for validated sample types. Antibody pairs are selected and tested to confirm the parallel detection of natural and recombinant standard protein and to ensure the accurate determination of target analytes within biological samples. Assays for each target analyte are screened against all target analytes to confirm low antibody cross-reactivity. |

Background
Pig animal models have an established track record of success in scientific research across a range of study areas, including cardiovascular, clinical work, pharmacology, toxicology, aging, and more recently in stem cell therapies. Porcine models share many similarities to human physiology, anatomy, and mechanisms of human disease, making them a unique valuable experimental model to test new disease therapies, devices and safety and efficacy of new drugs preceding human trials. In addition to transplantation research, a wide range of viral, bacterial, cancer, and inflammatory disease translational research has leveraged the pig’s close homology to humans. At the same time, some “minipig” breeds are providing unique value in longer-term pre-clinical studies due to their ability to age without growing too large, enabling researchers to complete multi-generational studies in comparatively compact timeframes. Emerging human disease research areas for porcine models include the fields of transgenics and regenerative medicine. For the near term, pigs are expected to remain an important translational model to bridge the divide between traditional rodent models and humans in advancing biological experimentation and improving human health.Assays for the Luminex platform are offered as High Performance Assays or Assays. The Luminex High Performance Assays are fully validated panels with a focused selection of analytes. They are available in We Mix, You Mix, and Predetermined formats. The Luminex Assays allows for the maximum number of analytes in a multiplex and is supplied as a premixed kit. View a table comparing the features and benefits of these bead-based multiplex assays.
Scientific Data
Porcine Luminex Discovery Assay Serum Sample Values Serum samples from 4 porcine were run on the Porcine Luminex Discovery Assay and sample values plotted. Minimum and maximum values shown by exterior lines, the box represents the 1st and 3rd quartiles with the center line representing the median.

FAQs
-
Are QC controls available for both the Luminex Discovery and High Performance Assays?
Controls are not offered for Luminex Discovery Assays, but may be formulated by the end user. A general guide may be available upon request. Controls are available for select Luminex High Performance Assays and can be found under the supplemental products tab on the product webpage. Some High Performance Luminex kits include controls and the controls will be listed in the materials provided section on the product datasheet. When included in the kit, controls are not sold separately.

Why does the Luminex Assay Customization Tool provide the option to split based on dilution?
The feature to split based on dilution enables investigators to configure premix Luminex panels based on sample type (cell culture supernates, serum, and plasma). This keeps analytes requiring the same dilution factor within a single panel, while separating analytes requiring different dilutions. The recommended dilution factor for each analyte may be different for each sample type, and the suggested sample dilutions are built into the Luminex Customization Tool. Note that the suggested dilution factors are based on samples from healthy volunteers.
-
Is the reconstituted standard a concentrate?
For Discovery Assays, each standard cocktail is a concentrate upon reconstitution with the volume specified on the Certificate of Analysis (CoA). After creating Standard 1 following the table in the product insert, the Standard 1 value is the concentration listed on the CoA.

At what frequency should Calibration and Verification tests be performed?
It is recommended that a Calibration test be performed within one week prior to running an assay. A Verification test should be performed prior to data acquisition on the day of running an assay.
