Recombinant Human TDP-43/TARDBP Protein, CF Summary

Carrier Free
CF stands for Carrier Free (CF). We typically add Bovine Serum Albumin (BSA) as a carrier protein to our recombinant proteins. Adding a carrier protein enhances protein stability, increases shelf-life, and allows the recombinant protein to be stored at a more dilute concentration. The carrier free version does not contain BSA.
In general, we advise purchasing the recombinant protein with BSA for use in cell or tissue culture, or as an ELISA standard. In contrast, the carrier free protein is recommended for applications, in which the presence of BSA could interfere.

Background: TDP-43/TARDBP
TDP-43 (TAR DNA binding protein of 43 kDa; gene name TARDBP) is a 414 amino acid (aa) nuclear RNA/DNA binding phosphoprotein that regulates transcription and splicing. TDP-43 is aberrantly expressed in most forms of amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration. In these cases it is absent from the nucleus and accumulates, along with a truncated 20-28 kDa form, in the Ubiquitin-containing inclusion bodies. TDP-43 can also influence CFTR exon 9 skipping in cystic fibrosis. Human TDP-43 is 96% identical to the mouse ortholog.

FAQs
-
Why is this protein supplied with 0.4 M urea in the formulation buffer?
Without the presence of urea, this protein would be insoluble. This is partially due to the expression system and the nature of the protein. Any attempt to dilute the urea down will cause the protein to precipitate.

Does the Recombinant Human TDP-43/TARDBP Protein (Catalog # AP-190-100) contain a tag?
AP-190 is the full-length protein and it does not contain any tags. The predicted molecular mass for this protein is 45 kDa, which matches the sequence stated in the datasheet linked here: https://www.ncbi.nlm.nih.gov/protein/NP_031401.1
