Detection of Hepassocin/FGL1 in Hepa 1‑6 Mouse Hepatoma Cell Line (Positive) and BaF3 Mouse Pro-B Cell Line (Negative) Cells. Hepassocin/FGL1 was detected in immersion fixed Hepa 1‑6 Mouse Hepatoma Cell Line (Positive) and BaF3 Mouse Pro-B Cell Line (Negative) Cells using Rabbit Anti-Mouse Hepassocin/FGL1 Monoclonal Antibody (Catalog # MAB11247) at 8 µg/mL for 3 hours at room temperature. Hepa 1-6 cells were treated with 3uM Monensin for 3 hours. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rabbit IgG Secondary Antibody (red; Catalog # NL004) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. View our protocol for Fluorescent ICC Staining of Cells on Coverslips.

Background:

Hepassocin, also known as hepatocyte-derived fibrinogen-related protein (HFREP-1) and Fibrinogen-like protein 1 (FGL-1), is a liver specific secreted protein containing one fibrinogen C-terminal domain. It exists as a homodimer and is reported to have hepatocyte mitogenic activity. Hepassocin/FGL-1 binds LAG3 independently from MHC class II (MHC-II) and acts as an Immune suppressive molecule that inhibits antigen-specific T-cell activation by acting as a major ligand of LAG3.
