Sample Values

Cell Culture Supernates - Human peripheral blood leukocytes were cultured in RPMI 1640 and supplemented with 10% fetal bovine serum. The cells were cultured unstimulated or stimulated with 10 μg/mL PHA for 2 or 5 days. Aliquots of the cell culture supernates were removed and assayed for levels of human MCP-1.

Product Summary

The Quantikine Human MCP-1 Immunoassay is a 3.5 or 4.5 hour solid phase ELISA designed to measure MCP-1 in cell culture supernates, serum, plasma, and urine. It contains E. coli-expressed recombinant human MCP-1 and antibodies raised against the recombinant factor. This immunoassay has been shown to accurately quantitate recombinant human MCP-1. Results obtained using natural human MCP-1 showed linear curves that were parallel to the standard curves obtained using the Quantikine kit standards. These results indicate that this kit can be used to determine relative mass values for natural human MCP-1.

Linearity

Background: CCL2/JE/MCP-1

CCL2/JE/MCP-1, is a chemokine that binds the receptor CCR2 and induces the chemoattraction of mononuclear cells. It induces the activation of monocytes, NK cells, lymphocytes, and basophils. Additionally, CCL2 promotes Th2 polarization in CD4+ T cells, and CCL2-mediated recruitment of monocytes to sites of inflammation contributes to disease severity in atherosclerosis, multiple sclerosis, and allergic asthma. Endogenous proteolytic trimming of CCL2 at the N-terminus, including the N-terminal pyrrolidone carboxylic acid-modified glutamine, downregulates activity but not receptor binding.

Assay Procedure

Refer to the product for complete assay procedure.

Bring all reagents and samples to room temperature before use. It is recommended that all samples, standards, and controls be assayed in duplicate.
  1.   Prepare all reagents, standard dilutions, and samples as directed in the product insert.
  2.   Remove excess microplate strips from the plate frame, return them to the foil pouch containing the desiccant pack, and reseal.

  3. 50 µL Assay Diluent for Serum & Plasma Samples Only
  4.   For Serum & Plasma Samples Only: Add 50 µL of Assay Diluent to each well.

  5. 200 µL Standard, Control, or Sample
  6.   Add 200 µL of Standard, control, or sample to each well. Cover with a plate sealer, and incubate at room temperature for 2 hours.
  7.   Aspirate each well and wash, repeating the process twice for a total of 3 washes.

  8. 200 µL Conjugate
  9.   Add 200 µL of Conjugate to each well.
  10.   For Serum & Plasma Samples: Cover with a new plate sealer, and incubate at room temperature for 2 hours.
    For Cell Culture Supernate & Urine Samples: Cover with a new plate sealer, and incubate at room temperature for 1 hour.
  11.   Aspirate and wash 3 times.

  12. 200 µL Substrate Solution
  13.   Add 200 µL Substrate Solution to each well. Incubate at room temperature for 30 minutes. PROTECT FROM LIGHT.

  14. 50 µL Stop Solution
  15. Add 50 µL of Stop Solution to each well. Read at 450 nm within 30 minutes. Set wavelength correction to 540 nm or 570 nm.

Urine was tested at 2-fold, 4-fold, and 8-fold dilutions. All samples were in range at 2-fold dilution, 75% were in range at 4-fold dilution, and 62.5% were in range at 8-fold dilution. All samples at all dilutions had a %Recovery within 80-120% of the reference (2-fold) dilution.

We continue to use this kit regularly.

Sample Tested: EDTA Plasma

MCP-1 concentrations were measured in plasma of HIV+ subjects on HAART and in healthy controls. Blood was collected in EDTA-tubes and plasma separated by centrifugation at 2000rpm for 20 minutes at 20oC. Plasma was diluted 1:2 before use in the assay.

Figure Legend for Figure:
a) Standard curve for MCP-1 from which values were extrapolated.
b) MCP-1 concentrations in plasma of HIV+ and HIV- subjects.

Values are expressed as pg/ml and are significantly elevated in HIV+?ART group. Analyses done using non parametric unpaired Mann Whitney U-test.
