Neuroscience Research Review: Neuronal EphB1 Activates a Neuroprotective Astrocyte Phenotype

Friday, December 15, 2017 - 08:36
Neuroprotective Astrocytes Induced by Neuronal EphB1

In the central nervous system (CNS), glia are key for maintaining tissue homeostasis. They function to support numerous processes including neurogenesis and neuronal survival, neural transmission, and immune surveillance. They also play a role in the initiation and progression of neuroinflammation and neurodegenerative disease. Astrocytes, the most abundant glial cell in the CNS, become activated in response to many insults, such as trauma, stroke, or neurodegenerative disease.1 Astrogliosis appears to be a complicated process as research is showing that the phenotype of reactive astrocytes may be defined by...

20 Years and Getting Stronger – The Ins and Outs of SUMOylation

Tuesday, December 12, 2017 - 16:35

Posttranslational modification with Small Ubiquitin-related Modifier (SUMO) proteins is now established as one of the key regulatory protein modifications in eukaryotic cells. Hundreds of proteins involved in processes such as chromatin organization, transcription, DNA repair, macromolecular assembly and protein homeostasis, trafficking and signal transduction are subject to reversible sumoylation. It is hence not surprising that disease links are beginning to emerge and that interference with sumoylation is being considered for intervention. In this webinar, Dr. Frauke Melchior provides an overview of fundamental mechanisms and highlight recent developments in the analysis and physiology of sumoylation.

RAS Oncoproteins: Therapeutic Vulnerabilities

Friday, December 1, 2017 - 14:05

Adrienne Cox, Ph.D. Associate Professor, UNC Chapel Hill

The three human RAS↗ genes (HRAS↗, KRAS↗ and NRAS↗) encode four highly related RAS small GTPases↗ (HRAS, KRAS4A, KRAS4B and NRAS). RAS proteins function as GDP-GTP regulated binary on-off switches that regulate diverse cytoplasmic signaling networks. In cancer and developmental disorders (RASopathies), mutationally activated RAS proteins drive aberrant signal transduction. RAS proteins are also the founding...

How to Keep Calm and Monitor Cell Stress, Death, and Behavior

How to Keep Calm and Monitor Cell Stress, Death, and Behavior

Measuring and monitoring cell health, stress, and behavior is a necessary part of using 2D and 3D cell culture models. This can be done qualitatively through microscopic assessment of cell structure, morphology, and confluency. Quantitative assessment of cell stress, death, and behavior is often needed when using 2D or 3D cell cultures for drug discovery, drug screening, and disease modeling. With the acquisition of Trevigen, Bio-Techne now has an extensive portfolio and unmatched expertise in assays and reagents for measuring cellular proliferation, viability, stress, DNA damage, and apoptosis. This blog showcases our expertise by highlighting the various resources and tools available through Bio-Techne.

CAR TCR Summit 2017 - Seaport World Trade Center

By Janice Frias, Ph.D., Bio-Techne Scientist

The CAR-TCR Summit in Boston from September 5-8 could not have had a better venue than the Seaport World Trade Center. The space was the perfect size for a group like this. It was able to accommodate all the concurrent talks and expo, yet keep the attendees in still a rather intimate setting to encourage interaction. And a glance out the window at the Boston Harbor in between talks was a nice perk.

The summit was very timely in that the week prior, the Novartis CAR T therapy, Kymriah, received FDA approval to fight leukemia in children and young adults. This was a hot topic, sparking speculation about who is...

Regulatory ILCs Suppress ILC1- and ILC3-Induced Innate Intestinal Inflammation

Regulatory ILCs Suppress ILC1- and ILC3-Induced Innate Intestinal Inflammation

Innate lymphoid cells (ILCs) are a recently described population of lymphocytes that play a critical role in innate immune responses and tissue remodeling. Multiple studies suggest that ILCs are particularly important for maintaining epithelial barriers and regulating immune responses at mucosal surfaces such as the lungs and intestines. Similar to T cells, ILCs consist of several subtypes that are characterized by the expression of different transcription factors and cytokines, and these correspond with the...

A Role for VSIG-3/VISTA Signaling in Acquired Resistance to Anti-PD-1 Therapy?

Wednesday, August 23, 2017 - 09:33
t-cell and tumor cell

Immunotherapy has revolutionized the way we treat cancer. Ground breaking studies have demonstrated that co-signaling molecules such as cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), programmed cell death 1 (PD-1) receptor, and programmed death ligand 1 (PD-L1) are an important therapeutic targets. However, these victories have been tempered by the development of acquired resistance in some cases. Why is this happening? What are the mechanisms of acquired resistance to immune checkpoint blockade?

Zika virus (ZIKV) is a mosquito-borne, single-stranded, positive-sense RNA virus belonging to the Flaviviridae family. It is closely related to other members of the Flavivirus genus including dengue virus, West Nile virus, yellow fever virus, tick-borne encephalitis virus, and Japanese encephalitis virus. ZIKV was first isolated from a rhesus monkey in Uganda in 1947 and the first human cases were detected in the 1950s in Uganda, Tanzania, and Nigeria. Since that time, there have been significant outbreaks in Africa, Southeast Asia, and the Pacific Islands, but the...
