M2a Microglia Activation State Markers

Overview

Microglia are the predominant resident immune cells in the brain. They become activated following exposure to pathological insults. Based on the activating stimulus encountered, reactive microglia can acquire different phenotypes that induce different effects, ranging from neuroinflammation to tissue reparation and neuronal protection. M2a microglia are the “alternatively” activated microglia that facilitate resolution of neuroinflammation. Microglia polarize to the M2a phenotype upon exposure to IL-4 or IL-13. These cells secrete IL-10 and various growth factors in order to attenuate the proinflammatory response and stimulate tissue repair, respectively.

Data Examples

ARG1 in Mouse M2 Microglia. Isolated mouse microglia were polarized to the M2 phenotype using Recombinant Mouse IL-4 (R&D Systems, Catalog # 404-ML). Arginase 1 (ARG1) was then detected in immersion-fixed mouse M2 microglia using a Sheep Anti-Human/Mouse/Rat Arginase 1/ARG1 Antigen Affinity-Purified Polyclonal Antibody (R&D Systems, Catalog # AF5868). These cells were stained using the NorthernLights™ 557-Conjugated Donkey Anti-Sheep IgG Secondary Antibody (R&D Systems, Catalog # NL010; yellow). Nuclei were counterstained with DAPI (blue). The cells were co-stained using a Rat Anti-Mouse CD11b/Integrin alpha M Monoclonal Antibody (R&D Systems, Catalog # MAB1124) and the NorthernLights™ 493-Conjugated Goat Anti-Rat IgG Secondary Antibody (R&D Systems, Catalog # NL015; green).
