5-HT1D in Human Brain. 5-HT1D was detected in immersion fixed paraffin-embedded sections of human brain (substantia nigra) using Mouse Anti-Human 5-HT1D Monoclonal Antibody (Catalog # MAB10354) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to neuronal processes. Detection of 5-HT1D in Rat Brain. 5-HT1D was detected in perfusion fixed paraffin-embedded sections of Rat Brain using Mouse Anti-Human 5-HT1D Monoclonal Antibody (Catalog # MAB10354) at 8 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using VisUCyte Antigen Retrieval Reagent-Basic (Catalog # VCTS021). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in neurons. Human 5-hydroxytryptamine receptor 1D, or 5-HT1D, is a G-protein coupled receptor for serotonin. 5-HT1D also functions as a receptor for ergot alkaloid derivatives, various anxiolytic and antidepressant drugs and other psychoactive substances. Ligand binding causes a conformation changes in a typical G-protein/adenylate cyclase fashion.
