Detection of Human Bcl‑10 by Western Blot. Western blot shows lysates of Raji human Burkitt's lymphoma cell line, A431 human epithelial carcinoma cell line, and Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 0.5 µg/mL of Goat Anti-Human Bcl-10 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2889) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for Bcl-10 at approximately 28 kDa (as indicated). This experiment was conducted using Immunoblot Buffer Group 4.
Bcl‑10 in A431 Human Cell Line. Bcl-10 was detected in immersion fixed A431 human epithelial carcinoma cell line using Goat Anti-Human Bcl-10 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2889) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Background: Bcl-10
B cell CLL/leukemia 10 (Bcl-10), a 233 amino acid, 26.2 kDa protein, is the cellular homolog of the equine herpesvirus-2 gene (E10). Bcl-10 is also known as CARD like apoptotic protein (CLAP), CED-3/ICH-1 prodomain homologous E10-like regulator (CIPER), celluar-E10 (c-E10), and CARD-containing molecule enhancing NF kappa B (CARMEN). Bcl-10 contains a caspase recruitment domain (CARD) and has been shown to induce apoptosis and to activate NF kappa B. Bcl-10 can interact with other CARD domain containing proteins including CARD9, 10, 11, and 14, which function as upstream regulators of NF kappa B signaling. Defects in Bcl‑10 may be involved in the pathogenesis of human malignancy.
