Applications

Recommended Concentration
Sample

Human CCL8/MCP-2 Sandwich Immunoassay

Recommended Concentration
Reagent
ELISA Capture (Matched Antibody Pair)
2-8 µg/mL

Use in combination with:

Detection Reagent: Human CCL8/MCP‑2 Biotinylated Antibody (Catalog # BAF281)

Standard: Recombinant Human CCL8/MCP-2 Protein (Catalog # 281-CP)

Neutralization
Measured by its ability to neutralize CCL8/MCP‑2-induced chemotaxis in the BaF3 mouse pro‑B cell line transfected with human CCR5. The Neutralization Dose (ND50) is typically 1-4 µg/mL in the presence of 0.25 µg/mL Recombinant Human CCL8/MCP‑2.

Please Note: Optimal dilutions should be determined by each laboratory for each application. General Protocols are available in the Technical Information section on our website.

Chemotaxis Induced by CCL8/MCP‑2 and Neutralization by Human CCL8/MCP‑2 Antibody. Recombinant Human CCL8/MCP-2 (Catalog # 281-CP) chemoattracts the BaF3 mouse pro-B cell line transfected with human CCR5 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog # AR002). Chemotaxis elicited by Recombinant Human CCL8/MCP-2 (0.25 µg/mL) is neutralized (green line) by increasing concentrations of Mouse Anti-Human CCL8/MCP-2 Monoclonal Antibody (Catalog # MAB281). The ND50 is typically 1-4 µg/mL.

Background: CCL8/MCP-2

CCL8, also known as Monocyte Chemoattractant Protein 2 (MCP-2), is an inflammatory CC chemokine that attracts monocytes, eosinophils and basophils. It is produced by many cell types and signals through interactions with CCR1, CCR2, CCR3 and CCR5.

Alternate Names
C-C motif chemokine 8; CCL8; chemokine (C-C motif) ligand 8; HC14; MCP2; MCP-2; member 8 (monocyte chemotactic protein 2); monocyte chemoattractant protein 2; SCYA10; small-inducible cytokine A8
