Detection of CD68/SR‑D1 in Human Macrophages by Flow Cytometry. Human CD14+ monocytes were selected from PBMCs using MagCellect Human CD14+ Cell Isolation Kit (MAGH105) and cultured for four days in recombinant human M-CSF (216-MC; 10 ng/mL) then stained with (A) Mouse Anti-Human CD68/SR-D1 Phycoerythrin-conjugated Monoclonal Antibody (Catalog # IC20401P) or (B) isotype control antibody (IC0041P) and Mouse anti-Human CD14 APC-conjugated Monoclonal Antibody (FAB3832A). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (FC005). Staining was performed using our Staining Intracellular Molecules protocol.

Background: CD68/SR-D1

CD68, also called Scavenger Receptor D1 (SR-D1), is a type I transmembrane glycoprotein that belongs to the LAMP family of molecules. CD68 is expressed on monocytes and macrophages and serves as a scavenger receptor for oxidized LDL.
