Western Blot Detection of Human Integrin a9 antibody by Western Blot. View Larger

Detection of Human Integrin alpha 9 by Western Blot. Western blot shows lysates of Tera2 human embryonic lung carcinoma cell line. PVDF membrane was probed with 2 µg/mL of Human Integrin a9 Monoclonal Antibody (Catalog # MAB4574) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for Integrin a9 at approximately 150 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Detection of Human Integrin alpha 9 by Western Blot Expression of VEGF-D-binding receptors by 468LN cells:(A) Expression of VEGF-R2 and R3 mRNA examined with semi quantitative RT-PCR, and (B) alpha 9 integrin mRNA measured with qRT-PCR. (C) Confirmation of VEGF-R2, VEGF-R3, and alpha 9 integrin expression at protein levels by western blot. MDA-MB-231 and MCF7-COX-2 cells were used as positive controls primarily for VEGF-R2 and VEGF-R3 expression. Differentially high alpha 9 integrin expression by 468 LN cells was evident. Background: Integrin alpha 9

Integrin alpha 9 is a 150 kDa type I transmembrane glycoprotein that is widely expressed and is found on smooth muscle, keratinocytes, skeletal muscle and hepatocytes. It principally associates with the beta 1 Integrin. alpha 9 beta 1 binds to VEGF-C and D, and osteopontin. The 951 amino acid (aa) extracellular domain of mouse Integrin  alpha 9 contains three beta -propellor repeats and multiple PheGly-GlyAlaPro repeats. A potential proteolytic cleavage site in human alpha 9 ECD is absolutely conserved in mouse alpha 9 ECD (Arg 566‑Val 567). The ECD of mouse Integrin alpha 9 shares 95% and 89% aa sequence identity to rat and human Integrin alpha 9, respectively.
