Detection of Human Jade‑1/PHF17 by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line and Ramos human Burkitt's lymphoma cell line. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human Jade‑1/PHF17 Monoclonal Antibody (Catalog # MAB6275) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for Jade‑1/PHF17 at approximately 92 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Jade‑1/PHF17 in HeLa Human Cell Line. Jade-1/PHF17 was detected in immersion fixed HeLa human cervical epithelial carcinoma cell line using Mouse Anti-Human Jade-1/PHF17 Monoclonal Antibody (Catalog # MAB6275) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Background: Jade-1/PHF17
Jade-1 (Gene for Apoptosis and Differentiation in Epithelia; also PHF17) is a 94 kDa member of the JADE family of proteins. It is expressed in renal tubule cells as well as embryonic primitive streak (in mouse). Initially, Jade-1 was described as a proapoptotic tumor suppressor. Jade-1 also interacts with the HBO1 complex to specifically acetylate nucleosome histones, thus acting as a transcriptional activator that likely impacts DNA replication, recombination and repair. Human Jade-1 is 842 amino acids (aa) in length. It contains one PEST domain with multiple Ser phosphorylation sites (aa 5-28), and two PHD Zinc fingers (aa 203-253 and 312-371) that are required for histone acetylation. There are two splice variants, one that is 61-64 kDa in size and contains a 7 aa substitution for aa 503-842, and another that shows a deletion of aa 99-110. Over aa 1-137, human Jade-1 shares 96% aa identity with mouse Jade-1.
