Background: Lysine Hydroxylase 2/PLOD2
Lysine Hydroxylase 2 (LH2) also known as PLOD2 (procollagen-lysine, 2 oxoglutarate 5 dioxygenase 2) is a membrane-bound homodimeric enzyme localized to the cisternae of the rough ER. The enzyme catalyzes the hydroxylation of lysine residues in collagen-like peptides, which in turn serves as sites for carbohydrate attachment in collagen. Two alternatively spliced forms have 737 or 758 amino acids (aa). Human LH2 (758 aa form) shares 92.5% aa sequence identity with the appropriate isoform of mouse and rat LH2.

western blot detection of PLOD2 in MDA-MB-231 transfected with siRNA against PLOD2. We routinely detect PLOD2 around 100KDa,this is likely related to post-translation modifications of PLOD2
