Detection of MMP-12 Catalytic Domain by Immunohistochemistry MMP-12 expression level becomes increasingly elevated as prostate cancer progresses: A, The MMP-12 protein expression (left) and quantification (right) in patients with pathological stages of benign prostatic hyperplasia (BPH), androgen-dependent prostate cancer (ADPC), and castration-resistant prostate cancer (CRPC) was determined by immunohistochemistry (scale bar 100 μm). B, The concentration of MMP-12 protein in the serum of the different groups of patients was determined by ELISA. Data are reported as means±SD. **P<0.01, ***P<0.001, and ****P<0.0001 unpaired t-test. ns: not significant. Background: MMP-12

MMP-12, also known as macrophage elastase, is a zinc and calcium dependent endopeptidase expressed constitutively in macrophages and can be induced in monocytes. MMP-12 can degrade a broad spectrum of substrates, including type IV collagen, fibronectin, laminin, vitronectin, proteoglycans, chondroitin sulfate, myelin basic protein, alpha 1-antitrypsin, and plasminogen. It can also activate MMP-2 and MMP-3. The MMP-12 precursor consists of a pro domain, a catalytic domain containing the zinc-binding site, and a C-terminal hemopexin-like domain.
