Background: IL-5
IL-5 is a disulfide-linked homodimeric cytokine that is secreted by T cells. IL-5 promotes the proliferation, differentation, and activation of eosinophils. It binds to a receptor complex consisting of one IL-5 specific alpha chain and one non-binding common beta chain that is shared with the receptors for GM-CSF and IL-3.

Splenocytes from C57BL/6 and Foxp3-deficient Scurfy mice were stimulated for 4 hours with phorbol 12,13-dibutyrate (500ng/ml), ionomycin (750ng/ml) and brefeldin A (2ug/ml). Cells were stained with fixable viability dye, then fixed with 2% formalin, permeabilized, and stained overnight for cytokines and gating markers (CD3, CD4).
Foxp3-deficient mice were chosen as a positive control because they are highly Th2-biased. IL-5 is not normally present in C57BL/6 CD4s in any great quantity. Another appropriate positive control would be ILC2 (e.g., gut lamina propria leukocytes).
Unstimulated cells that were otherwise treated identically served as a negative control.
