Detection of Human, Mouse, and Rat alpha -Cardiac Actin by Western Blot. Western blot shows lysates of human, mouse, and rat heart tissue. PVDF membrane was probed with 0.1 µg/mL of Mouse Anti-Human/Mouse/ Rat a-Cardiac Actin Monoclonal Antibody (Catalog # MAB9308) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for a-Cardiac Actin at approximately 42 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
alpha -Cardiac Actin in Human Cardiomyocytes. a-Cardiac Actin was detected in immersion fixed human induced pluripotent stem (iPS) cells differentiated to cardiomyocytes using Mouse Anti-Human/Mouse/Rat a-Cardiac Actin Monoclonal Antibody (Catalog # MAB9308) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Cells were fixated with methanol. Specific staining was localized to cytoplasm.
alpha -Cardiac Actin in Human Heart. a-Cardiac Actin was detected in immersion fixed paraffin-embedded sections of human heart using Mouse Anti-Human/Mouse/Rat a-Cardiac Actin Monoclonal Antibody (Catalog # MAB9308) at 1.7 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm of cardiomyocytes. Detection of Human, Mouse, and Rat alpha -Cardiac Actin by Simple WesternTM. Simple Western lane view shows lysates of human, mouse, and rat heart tissue, loaded at 0.2 mg/mL. A specific band was detected for alpha -Cardiac Actin at approximately 50 kDa (as indicated) using 2 µg/mL of Mouse Anti-Human/Mouse/Rat alpha -Cardiac Actin Monoclonal Antibody (Catalog # MAB9308). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: alpha-Cardiac Actin
Actin, alpha cardiac muscle 1 (ACTC1) is a 377 aa cytoskeletal filament in cardiac muscle which interacts with the 50-KDa domain of the myosin motor domain in cardiac muscle contraction. Actin is highly conserved across species, and differs from skeletal muscle actin (ACTA1) by only four amino acids. Mutations in the ACTC1 gene are linked to familial hypertrophy cardiomyopathy, atrial septal defects, arrhythmia, and chronic inflammatory cardiomyopathy.
