Detection of Human, Mouse, and Rat JAB1 by Western Blot. Western blot shows lysates of HepG2 human hepatocellular carcinoma cell line, PT18 mouse mast/basophil cell line, and L6 rat myoblast cell line. PVDF Membrane was probed with 1 µg/mL of Sheep Anti-Human/Mouse/Rat JAB1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF6390) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for JAB1 at approximately 38 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: JAB1

JAB1 (Jun activation domain binding protein-1), also called COPS5 or CSN5 (COP9 signalosome-5), is a 334 amino acid (aa), 38 kDa protein that is a subunit of the COP9 signalosome complex. It mediates cytosolic translocation of the cell cycle inhibitory protein, CDKN1B/p27Kip1, for degradation, thus promoting proliferation. It is widely expressed, especially in proliferating cells and up-regulates granulopoiesis when overexpressed. Within the region used as an immunogen, human JAB1 shares 99% aa identity with mouse and rat JAB1. A 379 aa isoform contains 93 alternate aa in place of the N-terminal 48 aa of the 334 aa form.
