Scientific Data
Detection of Human and Mouse SOCS‑3 by Western Blot. Western blot shows lysates of HeLa human cervical epithelial carcinoma cell line, Jurkat human acute T cell leukemia cell line, U937 human histiocytic lymphoma cell line, and C2C12 mouse myoblast cell line. PVDF membrane was probed with 0.1 µg/mL of Mouse Anti-Human/Mouse SOCS-3 Monoclonal Antibody (Catalog # MAB5696) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for SOCS-3 at approximately 30 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: SOCS-3
Suppressor of cytokine signaling 3 (SOCS-3), also known as CIS3, is a widely expressed 30 kDa protein that inhibits signal transduction induced by multiple cytokines including Erythropoietin, IL-6, LIF, Oncostatin M, and Leptin. It binds downstream signaling molecules such as JAK kinases and targets them for ubiquitin-mediated degradation, essential in a negative feedback control loop. Human SOCS-3 shares 96% aa sequence identity with mouse and rat SOCS-3.
