The Active STAT1 p91 DuoSet IC (R&D Systems, Catalog # DYC1490) can be converted to measure total STAT1 p91 using this antibody. Follow the protocol included with the kit, substituting this biotinylated antibody for the Biotinylated Double-stranded (ds) Oligonucleotide in step 1 of the Assay Procedure. 200 ng of STAT1 p91 antibody per binding reaction is recommended. For total STAT1 measurement, either whole cell extracts or nuclear extracts may be used.

Active vs. Total STAT1 p91 For total STAT1 p91 analysis, whole cell extracts were prepared from U937 human histiocytic lymphoma cells treated with recombinant human IFN-? (Catalog # 285-IF) for the indicated times. Extracts from 1 x 10⁶cells were incubated with 200 ng biotinylated goat anti-STAT1 p91 (replacing Biotinylated ds Oligonucleotide) for 30 minutes and analyzed in the Active Human/Mouse STAT1 p91 DuoSet IC Assay.For active STAT1 p91 analysis, nuclear cell extracts were prepared from U937 cells treated with recombinant human IFN-? for the indicated times, and assayed at 5 µg/well in the Active Human/Mouse STAT1 p91 DuoSet IC Assay.

Background: STAT1

STAT1 is associated with type I and II interferon signaling. Phosphorylation of STAT1 at Y701 leads to dimerization and translocation to the nucleus to activate gene transcription. Human STAT1 shows 93% and 94% aa identity with mouse and rat STAT1, respectively, over the region used as an immunogen. This region is identical between isoforms STAT1a (91 kDa) and STAT1b (84 kDa).
