Detection of Human Wnt‑4 by Western Blot. Western blot shows lysates of human breast cancer tissue. PVDF membrane was probed with 2 µg/mL Rat Anti-Human/Mouse Wnt-4 Monoclonal Antibody (Catalog # MAB4751) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (HAF005). A specific band for Wnt-4 was detected at approximately 39 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

PVDF membrane was probed with 2 µg/mL of Rat Anti-Human/Mouse Wnt‑4 Monoclonal Antibody (Catalog # MAB4751) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (HAF005). A specific band was detected for Wnt‑4 at approximately 39 kDa (as indicated). This experiment was conducted under reducing conditions and using Western Blot Buffer Group 1.
Wnt‑4 in MCF‑7 Human Cell Line. Wnt-4 was detected in immersion fixed MCF-7 human breast cancer cell line using Rat Anti-Human/Mouse Wnt-4 Monoclonal Antibody (Catalog # MAB4751) at 3 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rat IgG Secondary Antibody (red; NL013) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm. Wnt‑4 in Human Breast Cancer Tissue. Wnt-4 was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using Rat Anti-Human/Mouse Wnt-4 Monoclonal Antibody (Catalog # MAB4751) at 1.7 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Rat HRP-DAB Cell & Tissue Staining Kit (brown; CTS017) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in cancer cells. Wnt‑4 in Human Breast Cancer Tissue. Wnt-4 was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using Rat Anti-Human/Mouse Wnt-4 Monoclonal Antibody (Catalog # MAB4751) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Rat IgG VisUCyte™ HRP Polymer Antibody (VC005). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to plasma membrane. Human Wnt-4 ELISA Standard Curve. Recombinant Human Wnt-4 protein was serially diluted 2-fold and captured by Rat Anti-Human/Mouse Wnt-4 Monoclonal Antibody (Catalog # MAB4751) coated on a Clear Polystyrene Microplate (DY990). Sheep Anti-Human Wnt-4 Antigen Affinity-purified Polyclonal Antibody (AF6076) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (DY998) followed by Substrate Solution (DY999) and stopping the enzymatic reaction with Stop Solution (DY994).
