ORF73 HHV8 in Kaposi's Sarcoma Skin Tissue. ORF73 HHV8 was detected in immersion fixed paraffin-embedded sections of Kaposi's Sarcoma skin tissue using Mouse Anti-Human ORF73 HHV8 Monoclonal Antibody (Catalog # MAB10389) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Rat IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC005). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cell nuclei. View our protocol for IHC Staining with VisUCyte HRP Polymer Detection Reagents.

Background: ORF73 HHV8

Lana1 (also known as ORF73 or HHV8)  is a multifunctional protein that plays a role in the replication and long-term persistence of the viral episomal genome in dividing cells. Lana1 has been found to be associated with three different diseases observed in AIDS patients; kaposi's sarcoma, primary effusion lymphoma (which is a rare type of non-Hodgkin lymphoma affecting the body cavities) and multicentric Castleman's disease. To date there is much evidence to support a direct role for Lana1 in kaposi's sarcoma.
