Parkin in Human Brain. Parkin was detected in immersion fixed paraffin-embedded sections of human brain (substantia nigra) using Mouse Anti-Human Parkin Monoclonal Antibody (Catalog # MAB14382) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to neuronal cell bodies. Background: Parkin

Parkin is a 465 amino acid protein with an N-terminal ubiquitin-like domain, a linker region, a C-terminal TRIAD domain consisting of two RING finger motifs flanking a cysteine-rich IBR (in between RING fingers) motif. Mutations in Parkin are a major cause of autosomal recessive juvenile parkinsonism. Parkin-2 lacks exon 5, which encodes amino acid residues 179‑206 of Parkin.
