Scientific Data
Detection of Human Phospho-STAT4 (Y693) by Western Blot. Western blot shows lysates of NC-37 human Burkitt's lymphoma B lymphoblast cell line untreated (-) or treated (+) with pervanadate for 5 minutes. PVDF membrane was probed with 1 µg/mL Human Phospho-STAT4 (Y693) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4319) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band for Phospho-STAT4 (Y693) was detected at approximately 95 kDa (as indicated). For additional reference, duplicate lysates were probed with 1.0 µg/mL Human/Mouse STAT4 Polyclonal Antibody (lower panel, Catalog # PA-ST4). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: STAT4
Signal transduction and activator of transcription 4 (STAT4) is a member of the Jak/STAT signal transduction pathway. STAT4 mediates cytokine signaling by acting as a signal transducer in the cytoplasm and, upon phosphorylation, homo or heterodimerizes with other STAT proteins, which then translocate to the nucleus and activate transcription of specific genes. STAT4 is expressed in subset of tissues including spleen, heart, brain, peripheral blood cells and testis. STAT4 is involved in mediating IL-12 response in lymphocytes and regulating T helper cell differentiation.
