Detection of Human SLP‑76/LCP2 by Western Blot. Western blot shows lysates of THP-1 human acute monocytic leukemia cell line and Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 1 µg/mL of Human SLP-76/LCP2 Monoclonal Antibody (Catalog # MAB5637) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for SLP-76/LCP2 at approximately 90 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: SLP-76/LCP2

SLP-76 is a 533 amino acid (aa), 76 kDa tyrosine phosphorylated intracellular docking protein with a single SH2 recognition domain. It is expressed in T cells, platelets, neutrophils, NK and mast cells, and is required for progression of T cells to double positive stage in the thymus. Tyrosine phosphorylation is required for interaction with the signaling proteins VAV, NCK, PLC gamma, ITK, and ZAP70. SLP-76 may be phosphorylated by ZAP70 leading to NF-AT and IL2 gene activation. Human SLP-76 shares 83% and 85% aa sequence identity with mouse and rat SLP-76, respectively.
