Detection of Human SRPK3 by Western Blot. Western blot shows lysates of human skeletal muscle tissue. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human SRPK3 Monoclonal Antibody (Catalog # MAB72301) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for SRPK3 at approximately 59 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
SRPK3 in Human Skeletal Muscle. SRPK3 was detected in immersion fixed paraffin-embedded sections of human skeletal muscle using Mouse Anti-Human SRPK3 Monoclonal Antibody (Catalog # MAB72301) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Mouse HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS002) and counterstained with hematoxylin (blue). Background: SRPK3
SRPK3 (Ser/Arg-rich protein specific kinase 3), also called MSSK-1 (muscle-specific serine kinase 1) or STK23 (Ser/Thr kinase 23) is a 59 kDa (predicted) cytoplasmic member of the CMGC Ser/Thr protein kinase family. SRPK3 is principally expressed in heart and skeletal muscle. SRPKs selectively phosphorylate Ser on RS domain-containing proteins, initiating mRNA splicing and maturation. The 533 amino acid (aa) human SRPK3 contains a split kinase domain (aa 79‑292 and 347‑531) separated by a hinge region. A 491 aa isoform shows an alternate translation start site at aa 43. Within the region used as an immunogen, human SRPK3 shares 87% and 86% aa identity with mouse and rat SRPK3, respectively.
