Detection of Human STYK1 by Western Blot. Western blot shows lysates of SW480 human colorectal adenocarcinoma cell line, DLD clone 2C2 human colon adenocarcinoma cell line, and T47D human breast cancer cell line. PVDF Membrane was probed with 2 µg/mL of Mouse Anti-Human STYK1 Monoclonal Antibody (Catalog # MAB6668) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for STYK1 at approximately 48 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
STYK1 in PC‑3 Human Cell Line. STYK1 was detected in immersion fixed PC-3 human prostate cancer cell line using Mouse Anti-Human STYK1 Monoclonal Antibody (Catalog # MAB6668) at 10 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red, upper panel; Catalog # NL007) and counterstained with DAPI (blue, lower panel). Detection of STYK1 in HeLa Human Cell Line by Flow Cytometry. HeLa human cervical epithelial carcinoma cell line was stained with Mouse Anti-Human STYK1 Monoclonal Antibody (Catalog # MAB6668, filled histogram) or isotype control antibody (Catalog # MAB003, open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG F(ab')2Secondary Antibody (Catalog # F0102B).

Background: STYK1
STYK1 (Ser/Thr/Tyr kinase 1; also NOK, or “novel oncogene with kinase-domain”) is a 50 kDa, distant member of the FGFR/PDGFR family of tyrosine kinases. It appears to be intracellular, even though it contains a putative transmembrane segment. Both MAPK and PI3K pathways are activated by STYK1 activity, and STYK1 is associated with oncogenesis. Human STYK1 is 422 amino acids (aa) in length. It contains 25 aa N-terminus, a 21 aa putative transmembrane segment, and a 396 aa C-terminus that contains a Tyr-kinase domain (aa 118-372). Over aa 72-320, human STYK1 is 77% and 83% identical to mouse and canine STYK1, respectively.
