Detection of Human Tiki2/TRABD2B by Western Blot. Western blot shows lysates of HEK293 human embryonic kidney cell line either mock transfected or transfected with human Tiki2/TRABD2B. PVDF membrane was probed with 0.5 µg/mL of Sheep Anti-Human Tiki2/TRABD2B Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1600) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Tiki2/TRABD2B at approximately 65 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: Tiki2/TRABD2B
TIKI2, encoded by the TRABD2B gene, is a 517 aminoacid metalloproteinase that acts as a negative regulator of Wnt signaling by mediating the cleavage of N-terminal residues in many of Wnt proteins. Tiki matalloproteins are dependent on Mn2+/Co2+ and are inhibited by divalent metal chelators like EDTA. The other known Tiki protein in human, Tiki1, is required for head formation via Wnt cleavage-oxidation and inactivation, but lacks known metalloprotease motifs. TIKI2 is upregulated in renal cell carcinoma and it appears to suppress growth of osteosarcoma by targeting the canonical Wnt pathway
