Detection of TLR7 in RPMI8226 Human Cell Line by Flow Cytometry. RPMI8226 human myeloma cell line was stained with Mouse Anti-Human TLR7 Monoclonal Antibody (Catalog # MAB5875, filled histogram) or isotype control antibody (MAB003, open histogram), followed by Allophycocyanin-conjugated Anti-Mouse IgG F(ab')2Secondary Antibody (F0101B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (FC005). Staining was performed using our Staining Intracellular Molecules protocol.
Detection of TLR7 in Ramos Human Cell Line by Flow Cytometry. Ramos human Burkitt's lymphoma cell line was stained with Mouse Anti-Human TLR7 Monoclonal Antibody (Catalog # MAB5875, filled histogram) or isotype control antibody (Catalog # MAB003, open histogram), followed by Allophycocyanin-conjugated Anti-Mouse IgG F(ab')2Secondary Antibody (Catalog # F0101B). Background: TLR7
Toll-like receptor 7 (TLR7) is a 120 kDa (predicted, unglycosylated) type I transmembrane glycoprotein and member of the Toll-like receptor family. Human TLR7 is synthesized as a 1049 amino acid (aa) precursor that contains a 26 aa signal sequence, an 803 aa extracellular domain (ECD), a 21 aa transmembrane sequence, and a 189 aa cytoplasmic domain. Mature human TLR7 is 81% aa identical to mature mouse TLR7. TLR7 is detected in brain, placenta, spleen, stomach, small intestine, lung, and in plasmacytoid pre-dendritic cells. Functionally, TLR7 participates in the innate immune response to microbial agents.
