TNF RII/TNFRSF1B Specificity is Shown by Immunocytochemistry in Knockout Cell Line. TNF RII/TNFRSF1B was detected in immersion fixed THP‑1 human acute monocytic leukemia cell line but is not detected in TNF RII/TNFRSF1B knockout (KO) THP‑1 human cell line using Goat Anti-Human TNF RII/TNFRSF1B Antigen Affinity-purified Polyclonal Antibody (Catalog # AF726) at 5 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; NL001) and counterstained with DAPI (blue). Specific staining was localized to cell membrane. Staining was performed using our protocol for Fluorescent ICC Staining of Non-adherent Cells.
TNF RII/TNFRSF1B Inhibition of TNF‑ alpha -induced Cytotoxicity and Neutralization by Human TNF RII/TNFRSF1B Antibody. Recombinant Human TNF RII/TNFRSF1B Fc Chimera (726-R2) inhibits Recombinant Human TNF-a (210-TA) induced cytotoxicity in the L-929 mouse fibroblast cell line in a dose-dependent manner (orange line), as measured by Resazurin (AR002). Inhibition of Recombinant Human TNF-a (0.25 ng/mL) activity elicited by Recombinant Human TNF RII/TNFRSF1B Fc Chimera (0.075 µg/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human TNF RII/TNFRSF1B Antigen Affinity-purified Polyclonal Antibody (Catalog # AF726). The ND50 is typically 0.04-0.2 µg/mL in the presence of the metabolic inhibitor actinomycin D (1 µg/mL).
