Product Summary

The Quantikine™ Mouse IFN-alpha All Subtype immunoassay is a 3.5 hour solid-phase ELISA designed to measure IFN-alpha in mouse cell culture supernates, serum, and plasma. It contains HEK293-expressed recombinant mouse IFN-alpha and has been shown to accurately quantitate the recombinant factor. Results obtained using natural mouse IFN-alpha showed dose response curves that were parallel to the standard curves obtained using the Quantikine mouse kit standards. These results indicate that this kit can be used to determine relative mass values for natural mouse IFN-alpha.

Precision

Intra-Assay Precision (Precision within an assay) Three samples of known concentration were tested twenty times on one plate to assess intra-assay precision
Inter-Assay Precision (Precision between assays) Three samples of known concentration were tested in twenty separate assays to assess inter-assay precision. Assays were performed by at least three technicians using one lot of kit components

Recovery

The recovery of mouse IFN-alpha spiked to levels throughout the range of the assay in various matrices was evaluated.

Sample Type Average % Recovery Range %
Cell Culture Media (n=4) 98 91-106
EDTA Plasma (n=4) 105 91-121
Heparin Plasma (n=4) 106 96-120
Serum (n=4) 102 86-118

Linearity

To assess the linearity of the assay, samples containing and/or spiked with high concentrations of mouse IFN-alpha were serially diluted with calibrator diluent to produce samples with values within the dynamic range of the assay.

Scientific Data

Mouse IFN-alpha All Subtype Quantikine Natural Linearity Competitor Comparison IFN-alpha samples containing a high concentration in various matrices are diluted with appropriate Calibrator Diluent to produce sample values within the dynamic range of the assay. R&D Systems natural linearity is between 98%-103% compared to 125%-180% for the top competitor.

Mouse IFN-alpha All Subtype Quantikine ELISA Spiked Linearity Competitor Comparison IFN-alpha is spiked at high concentration in various matrices and diluted with appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay. R&D Systems linearity is between 88%-92% compared to 132%-201% for the top competitor.

Mouse IFN-alpha All Subtype Quantikine ELISA Spiked Recovery IFN-alpha is spiked at three known concentrations throughout the range of the assay and run to measure response of the spiked sample matrix. R&D Systems recovery is 99%.

Background: IFN-alpha

Interferon-alpha (IFN-alpha), also known as leukocyte interferon, represents a group of related but distinct proteins that share over 95% amino acid sequence homology. They are members of the type I interferon family which share a common cell surface receptor composed of two subunits, a 100 kDa ligand-binding subunit (IFN-alpha R2) and a 125 kDa ligand binding and signal transduction subunit (IFN-alpha R1) that is involved both in ligand binding and signal transduction. IFN-alpha has both anti-viral and immunomodulatory activities on target cells.
