Detection of Mouse and Rat PDZK1 by Western Blot. Western blot shows lysates of mouse liver tissue and rat liver tissue. PVDF Membrane was probed with 1 µg/mL of Goat Anti-Mouse/Rat PDZK1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5780) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for PDZK1 at approximately 75 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background: PDZK1

PDZK1 (PDZ Domain-containing 1; also CFTR-associated protein of 70 kDa/CAP70) is a 65‑70 kDa member of the NHER (Na/H Exchange Regulator) family of proteins. It is only expressed in epithelium, particularly that found in intestine, kidney and liver. PDZK1 serves as a scaffold protein that connects cell membrane proteins to cytoplasmic regulators. It also serves a critical role in basal-level bicarbonate secretion. Mouse PDZK1 is 519 amino acids (aa) in length and contains four PDZ domains (aa 9‑90; 128‑215; 243‑323; and 378‑458) plus three phosphorylation sites at Ser489, Ser492 and Ser514. Mouse PDZK1 shares 78% and 91% aa identity with human and rat PDZK1, respectively.
