Biological Activity
Pepstatin A Janelia Fluor® 526 is a fluorogenic green-emitting lysosome tracker and stain. No wash step is required when used for multicolor microscopy experiments. Pepstatin A Janelia Fluor® 526 can be used for live cell imaging, in confocal microscopy and super-resolution microscopy techniques including 3D-SIM (structured illumination microscopy) and STED, as well as two-color lattice light-sheet microscopy (lls). Excitation maximum = 530 nm, emission maximum = 549 nm.To measure the absorbance spectrum of this dye we recommend the following solvent: TFE plus 0.1% TFA.
Scientific Data
Application of Pepstatin A Janelia Fluor® 526 in live U2OS cells. Live U2OS cells were stained with Pepstatin A Janelia Fluor® 526 (2 µM) for 1 hour with Verapamil (10 µM) and imaged without an intermediate washing step on a LSM980 confocal microscope (Zeiss) using the following configuration: excitation: 514 nm / emission: 526–615 nm. Image kindly provided by Prof. Luke Lavis, Howard Hughes Medical Institute, Janelia Research Campus.
Application of Pepstatin A Janelia Fluor® 526 in fixed U2OS cells. Fixed U2OS cells were stained with Pepstatin A Janelia Fluor® 526 (2 µM) for 1 hour in 0.1 M sodium acetate buffer pH 4.5. Cells were imaged on a LSM980 confocal microscope (Zeiss) using the following configuration: excitation: 514 nm / emission: 526–615 nm.
