Detection of Protein A by Western Blot. Western blot shows Recombinant Protein A Long, Recombinant Protein A, and Recombinant Protein G (negative control). The left panel of the PVDF membrane was probed with 2 µg/mL of Rat Anti-Protein A Monoclonal Antibody (Catalog # MAB99761) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Protein A at approximately 33 kDa and Protein A Long at approximately 55 kDa (as indicated). To show specificity, the right panel was probed with Anti-Rat IgG Antibody (negative control). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Detection of Protein A by Simple WesternTM. Simple Western lane view recombinant Protein A Long, recombinant Protein A, and recombinant Protein G (negative control), loaded at 0.2 mg/mL. A specific band was detected for Protein A at approximately 40 kDa and Protein A Long at approximately 58 kDa (as indicated) using 20 µg/mL of Rat Anti-Protein A Monoclonal Antibody (Catalog # MAB99761) followed by 1:50 dilution of HRP-conjugated Anti-Rat IgG Secondary Antibody (HAF005). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Background: Protein A
The recombinant Protein A is a genetically engineered protein containing 5 IgG-binding regions of protein A. Cell wall binding region, cell membrane binding region and albumin binding region have been removed from the recombinant Protein A to ensure the maximum specific IgG binding. The recombinant Protein A is used in purification of polyclonal or monoclonal IgG antibodies. Protein A binds to various human, mouse and rat IgG subclasses (e.g., human IgG1, IgG2, IgG4; mouse IgG2a, IgG2b, IgG3; rat IgG2c). It also binds to total IgG from rabbit, guinea pig, pig, dog and cat.
